The seismic event occurred in Pulau Tanimbar at 6.20 am local time (2120GMT Friday), with the epicenter located 27 kilometers (16.7 miles) beneath the Earth's surface.
No immediate damage assessments have emerged from the affected region following the tremor.
The Saturday earthquake comes just one day after a separate 5.8-magnitude seismic event struck south of Indonesia's Java Island on Friday, resulting in structural damage to buildings and leaving 36 people injured.
Indonesia sits along the Pacific Ring of Fire, making it one of the world's most seismically active zones, where earthquakes and volcanic eruptions occur with regular frequency due to tectonic plate movements.
